There, my blessing with thee!: The "there" indicates that Polonius gives him a farewell blessing: a kiss? a hug? There is quite a bit of humor in the fact that after this farewell blessing, and after telling Laertes that he should already be aboard the ship, Polonius makes his son wait while he delivers one piece of advice after another.
